A boy and his beloved imaginary friend are able to stay together at an orphanage of sorts for imaginary friends that children have outgrown to be adopted by new children.

When 8 year old Mac's mother tells him that he's too old for an imaginary friend, he brings Blooregard "Bloo" Q. Kazoo to Foster's Home For Imaginary Friends where he is told that Bloo won't be adopted if Mac visits him every day. Soon after, Mac's older brother Terrence and an evil imaginary friend named Duchess make a plan to try to get rid of Bloo. Mac, with the help of friends named Wilt, Eduardo, and Coco, try to stop them.
Mr. Herriman sends Frankie to the mall to get streamers for Madame Foster's birthday party, and Mac, Bloo, Eduardo, Wilt, and Coco end up coming along. Eventually, they each wander off in a different direction after being distracted by something.
Bloo discovers a door in Foster's that everyone is forbidden to open, and nobody tells him what's behind it. One night he opens it, and releases Scribbles, a friend that does jobs around the house and causes everyone to become lazy.
After Bloo accidentally breaks a statue of Madame Foster, he and the other friends try to hide it from Mr. Herriman.
Bloo shows something to Mac from the roof of Foster's, and then neither can find their way downstairs for dinner. Mr. Herriman insists that no one can eat until Mac and Bloo arrive, and Madame Foster helps sneak them food while Herriman isn't looking.
Mr. Herrmian is filmed dancing embarrassingly by Mac and Bloo, and the video eventually ends up on the Internet which causes Herriman to be humiliated.
A new friend named Berry arrives at Foster's, and seems nice to everyone. Soon, she starts to try to kill Mac because she thinks that Mac is getting in the way of her being with Bloo, who she loves. Meanwhile, Bloo barely even realizes she's there.
Terrence creates a friend named Red to beat up Mac and Bloo; Bloo tries to get the same praise that Wilt does for rescuing imaginary friends, so he brings a man in a cell phone costume to Foster's.
Eduardo tries to hide a puppy he found from Mr. Herriman, who has a fear of dogs and doesn't let them in Foster's.
Mac and Bloo try to force the friends back in the house on Adopt-A-Thought Saturday so that they won't be adopted.
Mac and Bloo get sick after playing in the rain, causing them to miss watching a scary movie with the other friends. Wilt, Eduardo, and Coco mistake Bloo for a ghost, and Frankie gets locked out of the house in the rain.
Bloo plans to throw a wild party when Madame Foster and Frankie are away, but is afraid that Mac will ruin it by telling Mr. Herriman. Bloo gives Mac sugar to stop him from telling, but Mac becomes a sugar-crazed maniac.
Mac and the imaginary friends join Madame Foster's bowling team to help her beat her rival, Jerkins. Mac ends up getting kicked off the team and learning from a bowling expert, and the rival team promises Bloo a paddle ball if he joins them and helps them win.
When Wilt wants to watch a basketball game, he realizes that he can't say no to anyone's requests; A new imaginary friend named Bendy comes to Foster's and causes problems that everyone else is blamed for. Bloo tries to come up with a plan to prove Bendy is the one behind everything.
When a seeing eye friend named Ivan loses his blind kid, the friends help Ivan find him before something bad happens. After Mac takes Bloo to school for show and tell, the students create hundreds of Bloos. Mac then must determine who the real Bloo is before they go to the ice charades.
Bloo comes up with a plan to buy Foster's a new roof by selling Madame Foster's special, once-a-year cookies.
Mac and Bloo both develop a crush on Frankie, and try to stop her from having any contact with any guy. Eventually, they follow her on a date while in disguise.
Mac realizes that he accidentally created a new imaginary friend named Cheese. Cheese is allowed to stay at Foster's like Bloo, but Bloo has trouble dealing with his new brother Cheese.
At the arcade, the gang, sans Bloo rack up lots of tickets at all of its games. They win a rubber elephant, which Bloo grows to obsess about having. But when he swipes the elephant from Frankie, while she sleeps, he accidentally wrecks it and must replace it before everyone else finds out.
Bloo makes an offensive joke about an imaginary friend's height, and is told to meet this friend outside at 4:00.
Bloo pretends to be sick in order to appear on TV with the other friends. A Hollywood agent finds Bloo and turns him into a star by having him appear in deodorant advertisements. Soon, Bloo is not even allowed to see Mac.
Bloo discovers that Mac is a nerd, and tries to make him cool. When Mac is invited somewhere by the coolest kid in school and isn't at Foster's at 3:00, Mr. Herriman tries to get Bloo adopted.
Uncle Pockets, the very first friend to come to Foster's, returns. Bloo becomes jealous when Uncle Pockets gets all the attention, and tries to expose him as a fraud by using the house's newspaper.
A rich man is considering giving money to a charity, and wants to see if Foster's is worthy. Mac and Bloo are told to keep Coco out of the way. But Foster's is only considered for the money when Coco is mistaken as Mr. Herriman's wife. Mr. Herriman then tries to teach Coco how to be sophisticated.
Eduardo runs away to prove he's not a coward, and is found by Terrence, who plans to use Eduardo to win an imaginary friend battle.
Bloo decides to use his hiccups to win an imaginary friend talent show.
Madame Foster takes the gang camping, but Bloo ends up eating all of the food. Madame Foster then sends Mr. Herriman out into the woods to find something to eat, but things don't go well.
Someone named Goofball John McGee stumbles onto the Foster's property. Frankie thinks he's not really an imaginary friend, just a fraud; really a teenage kid in disguise, but everyone else believes otherwise. He always needs help with stuff, always at Frankie's expense. She is even more infuriated when they refuse to believe her when they think he really is an imaginary friend. So Frankie is more than anxious to expose him as a fake.
Duchess gets adopted by Mac's neighbors, but her constant loud complaining becomes too much for Mac's mother to handle. Terrence tells Mac that they're moving, and Mac tries to get his neighbors to get rid of Duchess. When this fails, he tries to bring her back to Foster's. Terrence warns everyone at the house, and they try to prevent her from getting back in.
Mac wins tickets for a trip to Europe, but everyone causes delays while packing. Already late for the trip, Frankie tries to make sure that Madame Foster can handle taking care of the house while she's gone, but Madame Foster just wants to break the rules.
Mac meets a girl named Goo who begins to follow him to Foster's every day. Her over-active imagination creates many new imaginary friends, which causes Foster's to become overcrowded.
Mr. Herriman's addiction to carrots causes him to hide carrots around the house. With no carrots, the only thing to have for dinner is a dish simply called "It." Mr. Herriman begins to punish anyone who comes near his hidden carrots, and Bloo tries to get in trouble so he doesn't have to eat "It."
Eduardo gets fleas, with whom he becomes close friends, much to the consternation of Frankie and Mr. Herriman. Bloo, on the other hand, wants some fleas of his own.
When several imaginary Santa Clauses are brought to Foster's, Mac begins to doubt that Santa is real. Bloo and the other friends try to prove to Mac that he exists. Bloo also tries to get Mr. Herriman to hand out more that one present to each person.
Mac creates a film about Foster's for a school project, but Bloo edits and it changes it into something completely different. Eventually, the film gets entered in a film festival.

The gang arrives at Mac's apartment for an unplanned slumber party on a school night, and Mac tries to keep them quiet so that they won't wake his mom.

After Frankie leaves the keys in the bus, Bloo takes Mac on a joyride and gets lost. Wilt promises to keep it a secret, but has trouble not telling anyone.
Foster's needs to get ready for a news show to promote themselves. The only problem is that Cheese keeps annoying everyone to the edge of insanity. So, Mr. Herrimen installs a new alarm system and messes up the code. Cheese is the only one who knows this code!
After Madame Foster rescues an imaginary friend, she is treated as a hero by everyone. Bloo also wants to be a hero so he founds his own home for imaginary friends. Mac tries to make Bloo stop since his home is nothing but a cardboard box and the only "imaginary friends" are two puppets that Bloo calls 'Purple' and 'Sharkhead'. Bloo mistakes a cat for an imaginary friend and makes a family adopt it. Soon Bloo remembers the great times he had with the cat and goes to the family's house to get it back.
Mac's teacher catches a student using an imaginary pen to cheat, and Mac and Bloo save him from the closet. At Foster's, the pen and a green ogre named Moose try to take over the house and turn it into a casino.
In her sleep, Goo accidentally creates all the villain characters from a make believe game that she had helped out on while the power was out at the home during a thunderstorm, who then imprison Mac and Bloo at the home. Now, the gang must finish the game to save their friends.
